In the beginning of the book you can tell that Lennie is unintentionally a big problem. Now you see this portrayed as soon as Lennie drinks the water without question.
Go forward, George gets mad at Lennie because he's bugging him for ketchup. Now, these are just small things. It is an ever growing thing. When they get to the ranch and Lennie gets them put on a watch list in the boss' eyes.
These are just the small things in the beginning of the book that foreshadow that something catastrophic would happen. It was one of the things that you never really thought about until you were nearing the end and started to expect new and worse things from Lennie.
It keeps going with Lennie looking at Curley's wife and you keep going on and he crushes Curly's hands. You can tell Lennie causes a lot of problems, regardless if he means to or not.
All of this leads up to the ending of the book. He kills the pup and then messes up as bad as you can by killing Curley's wife. This in turn makes the farm of George, Candy and Lennie dream almost impossible, and makes George not only lose his best friend but kill him.
Worst of all if you take a step back and look at all this you can see that nothing bad would have ever happened if Lennie were never there. This all goes to show that Lennie is the main antagonist of the story.
